Our lab studies how members of the ETS family of eukaryotic transcription factors are regulated by phosphorylation, sumoylation, autoinhibition and protein partnerships. One of these regulatory partners, Pax5, is a key player in cellular development and involved in certain types of cancer. The goal of my project is to use NMR spectroscopy to characterize the structural and dynamic basis for the interaction of Pax5 with DNA and Ets1. Particular emphasis will be on understanding how this partnership relieves Ets1 autoinhibition and thereby cooperatively controls the transcription of specific target genes.
